The reverse complement of a DNA sequence is formed by reversing the letters, interchanging A and T and interchanging C and G. Thus the reverse complement of ACCTGAG is CTCAGGT.

And DNA has two strands: Forward and reverse. Miseq is the technique to sequence the DNA for various purposes such as mutation detection, important for disease diagnosis. Hence, one need to find the mutations in both strands rather than only the forward to strand, to prevent false positives and other artifacts.
