1. During replication in bacteria there are chances that mutation had occurred. The mutation would cause genetic variation which would lead to natural selection. As due to mutation bacteria might get resistant to antibiotic say gene change in cell wall. There are chances that bacteria reside in biofilm that is deep seated and small amount would not be sufficient to penetrate and kill bacteria hence large dose will be required.
