Bovine intestinal alkaline phosphatase is an enzyme that normally operates in the intestines of cattle and has the function of breaking down a variety of phosphorylated compounds found in their food. Alkaline phosphatase can hydrolyze the artificial substrate p-nitrophenylphosphate (P-NPP). PNPP, which is colorless, turns yellow when de-phosphorylated by alkaline phosphatase to form the product, p-nitrophenol (P-NP). This reaction is summarized as follows: alkaline P-NPP (colorless) + H2O - P-NP (yellow) + inorganic phosphate phosphatase 1. You set up the following two tubes: Tube 1: 1mL of alkaline phosphatase + 4mL of 400 M P-NPP Tube 2: 1mL of alkaline phosphatase + 4mL of 200 M P-NPP Which tube would have a faster reaction rate? Explain your reasoning (why does concentration of p-NPP matter? Relate it back to week 4 lab concepts) 2. Alkaline phosphatase had an optimal temperature of 37°C. If you tested the enzyme at 37°C, 50°C, and 80°C, what would be your expected reaction rates? Graph your expected results (label your axes and lines). (1.5 point) Explain your reasoning for each line. 3. Enzyme X has an optimal pH that is basic. If you were to design an experiment to determine the optimal pH for this enzyme, which of the following pH range(s) would you test and why? a- pH 1-3 b- pH 3-5 c- pH 5-7 d- pH 7-9 e- PH 9-11 f- pH 11-14
